We present a constructive method to characterize topological properties and their connection to two-qubit quantum entanglement, in the framework of the tenfold classification and Wootters concurrence. This is established for the 2-qubit system through the antiunitary time reversal (TR) operator. Topological integers are computed systematically by a mapping to a quantum graph. The topological integers are measurable through geometric phase. This relation is governed by a sum rule, allowing a redistribution for implementations of a quantum gate using different Hamitlonians.
